D-Isocitric acid

چکیده
Mix**, wait for the end of the reaction (approx. 10 min) and read the absorbances of the solutions (A2). If the reaction has not stopped after 10 min, continue to read the absorbances at 2 min intervals until the absorbance increases constantly over 2 min. Determine the absorbance differences (A2-A1) for both, blank and sample. Subtract the absorbance difference of the blank from the absorbance difference of the sample.

The natural occurrence of optically active isocitric acid was first demonstrated by Nelson' and Bruce(2) in blackberry fruits, and a somewhat large accumulation of the acid was found by Pucher et al. 3~ in Bryophyllum leaf tissue. This acid has also been recognized as a member of the tri-carboxylic acid cycle in many animals, plants and microorganisms. All isocitric acid, found before in nature...
متن کاملThe preparation of optically active isocitric acid from Bryophyllum leaf tissue.
d-Isocitric acid’ has, in recent years, attracted much interest as one of the organic acids concerned in reactions catalyzed by the enzymes of various animal and plant tissues. A demand has accordingly arisen for specimens of this rare substance for use in investigations of the enzyme systems involved. The fortunate observation that d-isocitric acid is present in the leaves of succulent plants ...

Although several procedures have been described for the synthesis of isocitric acid, (l-4) the optically inactive product obtained may be contaminated with the diastereomeric alloisocitric acid and, if pure, contains at most only 50 per cent of the isomer of interest in biochemistry. Pucher, Abrahams, and Vickery (5) attempted to meet the need for this substance by devising a method for the iso...

Although citrate occupies a central position in the metabolic oxidation of carbohydrates, it is of considerable interest that at any one instant the largest proportion of total body citrate is found in the calcified tissues (1). Levels of citrate as high as 5.25 per cent of the dry, fat-free herring spine have been reported (2). The bone citrate is firmly bound, not being readily extracted with...

1. Oxidative dissimilation has been studied in enzymes from the honey bee. Using mitochondria isolated from the thoraces, complete oxidation of most of the TCA cycle members has been shown. 2. The presence of the acetate-activating enzyme, citrate-condensing enzyme, isocitric dehydrogenase, alpha-ketoglutarate dehydrogenase, glucose-6-phosphate, and 6-phosphogluconic dehydrogenase has been demo...

The biological conversion of d-isocitric to a-ketoglutaric acid was established by the work of Martius and others (1-4). Adler et al. showed that, with extracts of acetone powders of animal tissues, triphosphopyridine nucleotide (TPN) is a specific coenzyme in the reaction and cited evidence for a similar specificity in yeast. Ochoa (5) synthesized oxalosuccinic acid, the intermediate postulate...
